New device to detect Coronavirus strains from saliva sample within an hour

Researchers at the Massachusetts Institute of Technology and Harvard University have designed a small device called “miSHERLOCK” that can detect coronavirus strains from a saliva sample within an hour and as accurately as the PCR tests used now.
The device can also detect specific viral mutations associated with some of the currently circulating coronavirus mutations.
“In this study, targeting coronavirus mutations in the UK, South Africa and Brazil, you can easily adapt the diagnostic platform to address Delta variables and other emerging variables,” said James Collins, professor of medical engineering and science at the Massachusetts Institute for Medical Engineering and Science (IMES) and the Department of Biological Engineering. The device relies on CRISPR technology to detect the virus, which is a type of DNA sequences found in prokaryotes such as bacteria and ancient bacteria, and it is printed with 3D printing technology, and it costs only $ 15.
Reusing the device can bring the cost of individual tests down to $6 each, and that price may also drop significantly if the devices are mass-produced.
“miSherlock eliminates the need to transfer patient samples to a central test site and greatly simplifies sample preparation steps, giving patients and doctors a faster and more accurate picture of individual and community health, which is critical during the epidemic,” says Helena de Puig, a research associate from Wyss Institute and MIT.
Researchers point out that they have produced their new devices to make testing easier, simpler and more effective.
“The simple things that were ubiquitous in the hospital, such as nasopharyngeal swabs, became difficult to obtain due to the high demand for them, so routine sample processing procedures were disrupted, which is a big problem in a pandemic environment,” says Rose Lee, also a researcher at the Wyss Institute.
“Our team’s motivation for this project was to eliminate these hardships and provide accurate coronavirus diagnostics with less reliance on global supply chains, as well as accurately detect the variables that are emerging,” she added.

ويعتمد الجهاز على تقنية كريسبر (CRISPR) في اكتشاف الفيروس، التي تعد نوعا من تسلسلات الحمض النووي توجد في بدائيات النوى كالبكتيريا والبكتيريا القديمة، وهو مطبوع بتقنية الطباعة ثلاثية الأبعاد، ويكلف 15 دولارًا فقط.
ويمكن أن تؤدي إعادة استخدام الجهاز إلى خفض تكلفة الاختبارات الفردية إلى 6 دولارات لكل منها، وقد ينخفض هذا السعر أيضًا بشكل كبير إذا تم إنتاج الأجهزة على نطاق واسع.
وتقول هيلينا دي بويج، وهي عضو مشارك في البحث من معهد وايس (Wyss) ومعهد ماساتشوستس للتكنولوجيا “يلغي ماي شيرلوك الحاجة لنقل عينات المرضى إلى موقع اختبار مركزي، ويبسط إلى حد كبير خطوات تحضير العينة، مما يمنح المرضى والأطباء صورة أسرع وأكثر دقة لصحة الفرد والمجتمع، وهو أمر بالغ الأهمية أثناء تفشي الوباء”.
ويشير الباحثون إلى أنهم أنتجوا أجهزتهم الجديدة لجعل الاختبار أسهل وأبسط وأكثر فعالية.
وتقول روز لي الباحثة أيضا بمعهد وايس “الأشياء البسيطة التي كانت موجودة في كل مكان بالمستشفى، مثل المسحات الأنفية البلعومية، أصبح من الصعب الحصول عليها بسبب الطلب الشديد عليها، لذلك تعطلت إجراءات معالجة العينات الروتينية، وهي مشكلة كبيرة في بيئة الجائحة”.
وأضافت “كان دافع فريقنا لهذا المشروع القضاء على هذه الاختناقات وتقديم تشخيصات دقيقة لفيروس كورونا مع اعتماد أقل على سلاسل التوريد العالمية، وكذلك الكشف بدقة عن المتغيرات التي بدأت في الظهور”.
